Research context

Melanotan I (afamelanotide) is a synthetic tridecapeptide analogue of alpha-melanocyte-stimulating hormone (alpha-MSH), carrying norleucine and D-phenylalanine substitutions ([Nle4, D-Phe7]-alpha-MSH) that increase metabolic stability relative to the native hormone. It belongs to the melanocortin-agonist peptide class. In vitro and animal models describe it as an agonist ligand at melanocortin receptors, with reported selectivity toward MC1R.

Research in isolated melanocyte cultures and rodent models has examined melanotan I in relation to melanocortin-receptor binding, downstream cAMP/PKA signaling, and eumelanin-synthesis pathways including tyrosinase gene expression.
