Research context

SS-31 (elamipretide, MTP-131) is a synthetic, mitochondria-targeting aromatic-cationic tetrapeptide with the sequence D-Arg-2,6-dimethyl-Tyr-Lys-Phe-NH2. In vitro and in animal models it has been described as concentrating in the inner mitochondrial membrane through association with the phospholipid cardiolipin, where it has been studied for its influence on membrane structure and organization of the electron-transport machinery.

Studies in isolated cells and rodent models have examined SS-31 interactions with cardiolipin, mitochondrial cristae architecture, reactive-oxygen-species handling, and bioenergetic (oxidative phosphorylation) parameters. Analytical characterization of synthetic SS-31 preparations has been reported using reversed-phase HPLC for purity assessment and ESI-MS for confirmation of the tetrapeptide molecular weight.
