Research context

Tirzepatide is a synthetic 39–amino-acid peptide engineered as a dual agonist of the glucose-dependent insulinotropic polypeptide (GIP) receptor and the glucagon-like peptide-1 (GLP-1) receptor, two class B G-protein-coupled receptors. A fatty diacid moiety is incorporated to extend its circulating half-life relative to the native incretin peptides. In the published literature it has been characterized in vitro and in animal models as an agonist engaging both incretin receptor pathways.
